Abstract The Very High Gravity (VHG) fermentation is a technology that can lead to a reduction in waste generation, a reduction in energy consumption and GHG emissions and several technical, economic, and environmental advantages. Having, as a limiting factor, yeast tolerance to the most diverse stressors in the fermentation medium. To overcome this limitation, the aim of the work was to verify the potential protective effect of silica (+A) on Saccharomyces cerevisiae (Ethanol Red®) submitted to VHG fermentation. Initially, an adaptive test to VHG fermentation was carried out, with 5 cell recycles in musts from sugar cane syrup. Each recycle was subjected to the treatments, in quadruplicate: T1C (control) - Wort without silica supplementation; T2S100- Wort with supplementation of 100 mg L-1 of silica and T3S300- Wort with supplementation of 300 mg L -1 of silica. As a result, the T3S300 treatment in the adaptive test, showed viability of 77.5 to 81.55%; biomass production from 8.1 to 10.0 g L-1; yield from 90.0 to 95.3% and productivity from 7.3 to 10.9 mL L-1h-1. In conclusion, the treatment of the wort with silica (+A) (100 and 300 mg L-1) has an effect protector on yeast and may present positive responses in VHG fermentations.

O objetivo deste trabalho foi identificar isolados de fungos a partir de bagaço de cana-de-açúcar e madeira em decomposição e avaliar a sua atividade celulolítica em bagaço de cana. Cinco isolados foram avaliados, tendo-se como referências os fungos Trichoderma reesei QM9414 e T. reesei RUT C30. A atividade celulolítica foi estimada pela capacidade hidrolítica do extrato enzimático dos fungos cultivados em bagaço de cana sobre os substratos papel de filtro (atividade celulolítica total) e carboximetilcelulose sódica (atividade da endoglucanase). Os isolados foram identificados pela análise molecular da região 26S rDNA. Os gêneros Paecilomyces, Aspergillus, Acremonium/Penicillium e Trichoderma foram identificados. Embora T. reesei QM9414 tenha apresentado a mais alta atividade celulolítica total, alguns isolados também apresentaram alta atividade de endoglucanase. A biodiversidade, em nichos como bagaço de cana-de-açúcar, pode fornecer linhagens de fungos celulolíticos com grande potencial biotecnológico.

The objective of this work was to identify fungi isolates from sugarcane bagasse and decayed wood, and to evaluate their cellulolytic activity on sugarcane bagasse. Five isolates were evaluated, as compared to Trichoderma reesei QM9414 and T. reesei RUT C30. The cellulolytic activity was estimated through the hydrolytic capacity of the enzymatic extract of fungi grown on sugarcane bagasse over filter paper (total cellulolytic activity) and sodium carboxymethyl cellulose (endoglucanase activity). The isolates were identified by molecular analysis of 26S rDNA region. Paecilomyces, Aspergillus, Acremonium/Penicillium and Trichoderma genera were identified. Although T. reesei QM9414 showed the highest total cellulolytic activity, some isolates showed higher endoglucanase activities. Biodiversity found in habitats such as sugarcane bagasse can provide strains of cellulolytic fungi with great biotechnological potential.

Avaliaram-se no presente trabalho, as produções de etanol e dos ácidos acético e lático, bem como das proporções dos isômeros óticos D(-) e L(+) desse último, por 17 linhagens de Lactobacillus isoladas de fermentações industriais de produção de etanol. As linhagens foram crescidas a 32ºC por 24 horas, em meio contendo 1% de glucose, 1% de frutose, 1% de extrato de levedura, sais nutrientes (K, Mg e Mn) e tampão fosfato. Foram estimados os teores de ácido lático, ácido acético e etanol mediante cromatografia líquida de alta eficiência, assim como dos isômeros óticos D(-) e L(+) do ácido lático mediante espectrofotometria ao ultra-violeta, empregando desidrogenases láticas estereoespecíficas. O crescimento bacteriano foi inferido pela absorvância a 600 nm. Os resultados obtidos mostraram, pelos perfis de excreção dos metabólitos, a presença de 8 linhagens homofermentativas obrigatórias (produzindo unicamente ácido lático), 8 linhagens heterofermentativas obrigatórias (com produções de ácidos lático, acético e etanol) e 1 linhagem supostamente heterofermentativa facultativa. Observou-se também, em relação à formação dos estereoisômeros, que 12 linhagens foram incluídas no grupo DL, 4 no grupo L e 1 no grupo D. Os resultados permitem concluir que os Lactobacillus que contaminam processos fermentativos industriais de produção de etanol, podem se apresentar nos 3 biotipos fermentativos e produzindo as mais variadas proporções dos dois estereoisômeros do ácido lático, com relevantes implicações biotecnológicas. Este é o primeiro relato sobre as produções dos isômeros óticos do ácido lático por bactérias do gênero Lactobacillus isoladas de fermentações industriais baseadas na cana-de-açúcar.

The aim of the present work was to evaluate the metabolism type of 17 Lactobacillus strains isolated from industrial ethanol fermentation plants. The strains were grown at 32°C for 24 hours on a mixture of equal amounts of glucose and fructose as the carbon source, and supplemented with yeast extract, mineral nutrients and buffer. Bacterial growth was estimated by absorbance at 600nm and the main end products of bacterial metabolism (lactate, acetate and ethanol) were measured by high performance liquid chromatography, while the stereoisomers, D(-)- and L(+)-lactate, were assayed by an enzymatic methodology using stereospecific lactate-dehydrogenases. According to the results, all the three types of metabolism were found among the bacteria investigated: obligately homofermentative (8 strains), facultative heterofermentative (1 strain) and obligately heterofermentative (8 strains). The results have showed a predominance of DL strains regarding the stereoisomers production, but it was also found strains producing a single type of the isomeric form. These findings suggest the possibility to explore the lactobacilli biodiversity in fuel ethanol fermentation plants for lactate production of chemically pure optical isomers. This is the first report on lactic acid isomers formation by Lactobacillus strains isolated from sugar cane based-industrial fermentations.

O presente trabalho teve por finalidade estudar os efeitos do cádmio sobre a levedura Saccharomyces cerevisiae, bem como avaliar a possibilidade de se utilizar a vinhaça como fornecedora de agentes ligantes, visando minimizar os efeitos deletérios do mesmo. Primeiramente montou-se um ensaio visando observar a ação tóxica de diferentes concentrações de cádmio (0; 0,05; 0,1 e 0,5mM), avaliada pelo crescimento de duas cepas da levedura S. cerevisiae (PE-2 e IZ-1904) em meio YED. O meio foi inoculado com 1mL de uma suspensão a 1% (m/v) das respectivas cepas e incubado por 18 horas. Em tempos determinados durante o crescimento anaeróbio, alíquotas da suspensão de células foram retiradas e a concentração celular foi determinada. No final do ensaio, foram determinadas a viabilidade celular, a taxa de brotamento e a contaminação bacteriana. Os teores de trealose para cada tratamento, de ambas as cepas, foram dosados no início e no final do ensaio. Em uma segunda etapa, montou-se um ensaio visando avaliar a capacidade da vinhaça (0,15 e 30% do volume do meio) em atenuar os efeitos tóxicos de duas doses de cádmio (0,1 e 0,5mM), empregando-se a levedura S. cerevisiae PE-2 em meio YED. O meio foi inoculado com 2mL de uma suspensão a 1% (m/v) da levedura e incubado por 18 horas. Em tempos determinados durante o crescimento anaeróbio, alíquotas da suspensão de células foram retiradas e a concentração celular foi determinada. No final do ensaio, foram determinadas a viabilidade celular, a taxa de brotamento, a contaminação bacteriana e a produção de etanol. Os teores de trealose, para cada tratamento, foram dosados nas leveduras no início e no final do ensaio. O cádmio prejudicou o crescimento e a viabilidade celular das duas cepas da levedura S. cerevisiae. A vinhaça apresentou um discreto efeito tóxico, traduzido pela redução do crescimento. Porém, nos tratamentos contaminados com cádmio, apresentou um efeito protetor, minimizando os efeitos deletérios do metal.

The present study was carried out in order to evaluate the capability of different cadmium concentration (0; 0,05, 0,10 and 0,50mM) to affect the growth of two S. cerevisiae strains (PE-2 and IZ-1904) in YED (yeast extract 1% and dextrose 2%) medium, and to evaluate the three vinasse concentration capability (0,15 and 30%) to attenuate the two cadmium concentration toxicity (0,1 and 0,5mM), using S. cerevisiae PE-2 strain in YED medium. In the first assay, the medium was inoculated in aseptic conditions with 1mL of 1% yeast suspension (PE-2 or IZ-1904) and incubated at 30ºC, 70 RPM for 18 hours. During anaerobic growth (0, 2, 4, 6, 8, 10, 12, 14, 16 and 18 hours), portions of cell suspension were taken out and biomass concentration was determined. At the end of fermentation, yeast viability, budding rate and bacterial contamination were determined. Both, initial and final trehalose, was measured. In the second assay, the medium was inoculated in aseptic conditions with 2mL of 1% PE-2 suspension and incubated at 30ºC, 120 rpm for 18 hours. During the anaerobic growth (0, 2, 4, 6, 8, 10, 12,14, 16 and 18 hours) portions of cell suspension were taken out and biomass concentration was determined. At the end of fermentation, alcohol production, yeast viability, budding rate and bacterial contamination were determined. Both, initial and final trehalose, was measured. The increase of cadmium levels showed a reduction on yeast growth and cell viability. Vinasse showed low toxicity, but protected yeast cells very effectively against the toxic effects of cadmium.

A adubação fosfatada corresponde à fração mais onerosa do custo de produção da cultura da soja. A obtenção de cultivares de soja eficientes na absorção e utilização de fósforo (P) em condição de média disponibilidade deste nutriente pode contribuir para aumentar o potencial produtivo da cultura. Trinta e dois cultivares de soja [Glycine max (L.) Merr.], de ciclo precoce, semiprecoce, semitardio e tardio, recomendados para o cerrado, foram cultivados em Latossolo Vermelho-Amarelo distrófico típico, do cerrado, objetivando avaliar a atividade da fosfatase ácida, concentração de P na folha diagnóstico e biomassa da parte aérea. Ocorreram diferenças entre os cultivares dentro de todos os ciclos de maturação na atividade da fosfatase ácida e na biomassa da parte aérea. Cultivares de ciclos semitardio e tardio apresentaram diferenças significativas quanto à concentração de fósforo na folha diagnóstico. A atividade da fosfatase ácida correlacionou-se positivamente com a biomassa da parte aérea nos cultivares dos ciclos semiprecoce (r = 0,46) e tardio (r = 0,47) e, negativamente (r = -0,40), com a concentração de P na folha-diagnóstico, nos cultivares do ciclo tardio. A ocorrência de cultivares de soja com alta e baixa atividade da fosfatase ácida dentro do mesmo ciclo de maturação sinalizam a existência de diferentes mecanismos envolvidos na mobilização de P no solo e remobilização interna deste nutriente na planta entre os grupos de cultivares.

The phosphate fertilization represents the most costly fraction of soybean crop production. Efficient soybean cultivars for P absorption and utilization in soils of medium available P are highly desirable and might contribute for increasing crop production potential. Thirty two soybean [Glycine max (L.) Merr.] cultivars recommended for 'Cerrado' and differing in growth cycle (early, semi-early, semi-late, and late) were grown in a dystrophic Typic Haplustox Cerrado soil to evaluate the acid phosphatase activity, P content in the diagnostic leaf, and shoot biomass. There were differences among the soybean cultivars within all maturation groups in acid phosphatase activity and shoot biomass. The diagnostic-leaf P-content showed significant differences on semi-late and late maturation groups' cultivars. The acid phosphatase activity correlated positively with the plant shoot biomass from semi-early (r = 0.46) and late (r = 0.47) cultivars, and negatively (r = -0.40) with the P content in the diagnostic leaf of late maturation cultivars. The occurrence of soybean cultivars with high and low acid phosphatase activity within the same maturation groups indicates the existence of different mechanisms involving P mobilization in the soil and internal plant P remobilization.

A fermentação dos carboidratos de reserva, glicogênio e trealose é um procedimento para aumentar o nível de proteína das células de leveduras com simultâneo aumento na produção de etanol. Este trabalho estudou a cinética de degradação do glicogênio e trealose em duas linhagens industriais de Saccharomyces cerevisiae (PE-2 e SA-1), bem como o efeito de diferentes temperaturas (38º, 40º, 42º e 44ºC) na velocidade de degradação. A fermentação endógena foi conduzida com suspensão de leveduras a 20% (m/v) em massa úmida, no vinho com 3 a 4,5% (v/v) de etanol. A degradação dos carboidratos de reserva, a 40ºC, seguiu uma cinética de primeira ordem, mostrando que sua taxa é dependente da concentração dos carboidratos na célula. A taxa especifica de degradação (k) variou de 0,0387 a 0,0746 h-1. Em relação a outros parâmetros analisados a 40ºC, foi observado que a viabilidade e biomassa seca e úmida foram reduzidas, enquanto a reserva de proteína celular e etanol, glicerol e nitrogênio no meio aumentaram. A degradação do glicogênio e trealose em diferentes temperaturas (38ºC, 40ºC, 42ºC e 44ºC) mostrou que a 38ºC a taxa de degradação foi a menor, ao passo que a partir de 42ºC ou superior, a degradação do glicogênio não mais progrediu após poucas horas de incubação. Portanto, do ponto de vista prático, a melhor temperatura de incubação é em torno de 40ºC. A aplicação da equação de Arrheniusmostrou que as energias de ativação de 40ºC a 42ºC foram 165,90 e 107,94 kcal.ºK-1.mol-1 para trealose e glicogênio respectivamente para a linhagem PE-2, e 190,64 e 149,87 kcal.ºK-1/mol-1, para a linhagem SA-1 respectivamente.

The fermentation of yeast reserve carbohydrates, glycogen and trehalose is a procedure to increase protein level of yeast cells and improve ethanol production. This work studied on the degradation kinetics of glycogen and trehalose carried out with two industrial strains of Saccharomyces cerevisiae (PE-2 and SA-1) and the effect of different temperatures (38º, 40º, 42º and 44ºC) on degradation rate. Endogenous fermentation was carried out in a yeast suspension at 20% (w/v) based on wet weight, suspended in fermented media with 3.0 to 4.5 % (v/v) of ethanol. The degradation of the carbohydrate reserves at 40ºC followed first-order kinetics, showing that its rate is mainly dependent on the carbohydrate concentration in the cell. The degradation rate (k) ranged from 0.0387 to 0.0746 h-1. Analyzing other parameters at 40ºC, it was observed that viability and dry and wet yeast biomass were reduced while cell protein, ethanol, glycerol and nitrogen in the medium increased. Glycogen and trehalose degradation at different temperatures (38º, 40º, 42º e 44ºC) showed that at 38ºC the degradation rate was slow and from 42ºC on the degradation of glycogen stopped after few hours of incubation. Thus, from a practical point of view, the best incubation temperature is around 40ºC. The application of the Arrhenius equation showed that activation energy from 40º to 42ºC was 165.90 and 107.94 kcal.ºK-1.mol-1 for trehalose and glycogen respectively for PE-2 strain, and 190.64 and 149.87 kcal.ºK-1.mol-1 respectively for SA-1 strain.
